It wasn't a full spanking, but Lisa Kudrow's character (Phoebe) did give Jennifer Aniston as Rachel a good pop on the behind in an episode of Friends, it was actually used in the opening montage for a while.

There was a nice spanking threat in Dr Who a year or two back. I think the episode was called 'Dinosaurs on a Spaceship', and it featured Queen Nefertiti and an Edwardian big-game hunter as subsidiary characters. And at one point, when QN was being especially capricious, the hunter explicitly threatened her with a spanking. Alas, he didn't follow through; but the Doctor told them to stop flirting.
