For the life of me I can't work out how you reply to a thread with a part of someones text and the green vertical line. Can anyone help me please?

Highlight their text, then click on quote.

Highlight the text you want to quote and then click Quote from the top of the post you are quoting from...

To do what i've just done above, you first select the text you want to use, then click on "Quote" which copies the text to the editing box, then you can add your own message below it.
